1. Piperine Overview

Piperine is main active component of pepper. It is a type of alkaloid. It is found widely in nature, especially in pepper plants. Piperine accounts for 4%-9% of pepper fruit. Plamed Piperine comes from dried fruits of black pepper. It is 95% purity, and acts as a natural cosmetic ingredient. Piperine is light yellow fine powder. It is extremely sensitive to light. It is almost insoluble in water, and slightly soluble in ether; it is soluble in benzene or acetic acid, and easily soluble in chloroform, ethanol.

In recent years, with the research in cosmetics field, it has been found that Piperine can promote skin penetration. It is responsible for enhancing effective ingredients of cosmetics to quickly penetrate into skin. Therefore, it can be added into cosmetics as osmotic agent. Pepper (Latin name: Piper nigrum L) is a wooden climbing vine of Piperaceae family. It is native to southeast Asia. Pepper is extensively distributed in tropical areas, such as Indonesia, India, Malaysia, Sri Lanka and Brazil. Pepper in China mainly grows in Taiwan, Fujian, Guangdong, Guangxi and Yunnan provinces. Pepper can be divided into black pepper and white pepper. The cooked and dried unripe fruits are black pepper, when harvested in late autumn to the second spring. The fruit is harvested when it turns red; then soak with water for a few days, and wipe away the flesh, dry and then obtain white pepper. Black pepper is more pungent than white pepper. White pepper is of great medicinal value; it can dispel cold and be good for stomach. Cosmetic uses dried fruits of black pepper. Pepper fruit contains a variety of amide compounds, mainly including piperine. It also includes volatile oil ingredients, such as piperonal, dihydrocarveol, caryophyllene oxide, and sabinene. At present, extracts from all parts of pepper are widely applied in medicine, food, health care products and cosmetics.

• Anti-inflammation

Piperine can inhibit production of pro-inflammatory cytokines. Thereby, it cuts down expression of inflammatory cytokines such as interleukin 1, interleukin 6, tumor necrosis factor α, etc. Therefore, Piperine can reduce inflammation and pain. It exerts an anti-inflammatory effect.

• Promotion of penetration

Piperine can be absorbed through skin, and act directly on subcutaneous tissues, nerves and blood vessels. Thus Piperine can significantly improve and promote absorption and utilization of active ingredients in skin. Also it can help cosmetic ingredients quickly penetrate into skin.

• Anti-oxidation

Piperine can reduce oxidative stress damage of cells in the process of oxygen free radicals generation and scavenging. Additionally, it has certain scavenging capacity to hydroxyl and DPPH free radicals. It exhibits anti-oxidation property. Its anti-oxidative capacity is equivalent to 60.05% of vitamin C; and it is stable in acidic medium.

• Other function

Piperine can stimulate activity of melanocytes. It increases the amount of tyrosine, and promotes expression of TPR1. Thus Piperine accelerates synthesis of melanin in epidermal melanocytes. It can be added into cosmeceuticals for treatment of depigmentation dermatosis.

6. Piperine Stability

• Photo-stability

Put certain concentration Piperine ethanol solution in natural light. Measure regularly its absorbance at 344 nm under respectively 0, 1, 2, 3, 4, 5 hour of sun exposure. The result can be seen from table 1. The absorbance decreases with illumination time. But after 5 hours natural light, absorbance only drops by 1.3%. It indicates that chromogenic groups in Piperine’s chemical structure change little.

• Effect of different pH values on Piperine

Use diluted NaOH solution and HCL solution to adjust pH value of Piperine w=95% ethanol solution. And then measure absorbance of the solution under different pH media. The result shows in table 2 that Piperine is stable in acidic medium, but the absorbance value increases in alkaline medium.

• Heat stability

Put a certain concentration Piperine w=95% ethanol solution in 60℃ constant temperature water for processing. Keep warm for separately for 0, 1, 2, 3, 4, 5 hours. Measure the absorbance after cooling to room temperature. Table 3 shows that after 2h 60℃ insulation treatment, absorbance value drops by 6.4%. But then absorbance value barely has change.
