Plamed has launched a new natural cosmetic material PMRepair® Centella, it has excellent skin barrier repair and anti-inflammatory effects.
PMRepair® Centella is composed of Asiaticoside, Madecassoside, And Centella Asiatica Flower/Leaf/Stem Extract. It’s mainly for skin barrier repair. Plamed has proved its effect for skin repair.
- PMRepair®Centella for skin repair principle
First, the anionic surfactant sodium lauryl sulfate (SLS) is used to stimulate the 3D epidermal skin model (EpiKutis®) to construct an in vitro skin injury model. Changes in filaggrin (FLG), loricrin (LOR) and transglutaminase 1 (TGM1) content are used to evaluate the repair effect of the tested samples.
Test concentration: the efficacy test concentration of asiaticoside (95% purity) is 0.5mg/mL, and that of centella triterpenes is 0.15mg/mL. After testing, both the tested samples do not show obvious cytotoxicity based on keratinocytes at the concentration of 0.5mg/mL and 0.15mg/mL respectively.

Compared with NC group, the living cells of the basal layer in sample group PMRepair® Centella increased, the cells arrangement was close, and the vacuoles phenomenon was improved. The living cell layer in sample group Asiaticoside (95%) was thinner, the vacuoles phenomenon was not improved, and the stratum corneum was loose and thickened.

In PMRepair® Centella group, the number of living cells increases and the stratum corneum is tight, and the thickness of the living cell layer increases significantly, with an increase rate of 49.39%.
- PMRepair®Centella for skin repair in filaggrin (FLG)
FLG (Filaggrin) is an important component of keratin envelope, which can coalesce keratin into bundles; it can also be hydrolyzed into natural moisturizing factors.

Compared with NC group, the FLG contents in sample groups PMRepair® Centella and Asiaticoside (95%) increased significantly, with the improvement rates of 150.00%, 45.24%, respectively.
- PMRepair®Centella for skin repair in Loricrin (LOR)
LOR (Loricrin) accounts for more than 70% of the keratin envelope, reinforcing the barrier function of the epidermis, and its reduced content is a major factor in the weakening of the skin barrier function.

Compared with NC group, the LOR content in sample group PMRepair® Centella increased significantly, with the improvement rate of 646.15%. The LOR content in sample group Asiaticoside (95%) decreased significantly.
- PMRepair®Centella for skin repair in transglutaminase 1 (TGM1)
TGM1 is able to transfer amino acids to glutamate residues of proteins to form isopeptide bonds, which is key step in the formation of keratin envelope.

Compared with NC group, the TGM1 content in sample group PMRepair® Centella increased significantly, with the improvement rate of 537.14%. The TGM1 content in sample group Asiaticoside (95%) did not change significantly.
In conclusion, based on the 3D epidermal skin model (EpiKutis®), compared with the control group:
The tissue morphology of the sample PMRepair® Centella group was significantly improved at 0.15% concentration, the thickness of the living cell layer was significantly increased by 49.39%, and the content of filaggrin (FLG) , loricrin (LOR) and transglutaminase 1 (TGM1) were significantly increased by 150.00%, 646.15% and 537.14% respectively. It was considered that 0.15% of PMRepair® Centella achieved skin repairing effects by improving tissue morphology, promoting skin tissue remodeling and enhancing the content of barrier-related proteins (FLG, LOR, TGM1) to strengthen the barrier.
However, Asiaticoside (95%) at 0.5% concentration showed no significant improvement in tissue morphology and no significant change in the thickness of living cell layer, the content of filaggrin (FLG) increased by 45.24%, the content of loricrin (LOR) decreased significantly, and the content of transglutaminase 1 (TGM1) showed no significant change.
Therefore, PMRepair® Centella is cost-effective in skin repair, 9 times that of asiaticoside. Its INCI name is composed of ASIATICOSIDE, MADECASSOSIDE, CENTELLA ASIATICA FLOWER/LEAF/STEM EXTRACT.
It’s better to dissolve in 35% butanediol & 65% deionized water; the solution is clarified and transparent after a long time.
