1. PMSoothe® 6521 Overview
PMSoothe® 6521 is a combination of Kava Pepper Root Extract and Gentian Root Extract. Piper methysticum Forst is a perennial evergreen shrub-like medicinal plant of the Piperaceae family; it is widely used in nutritional supplements and herbal preparations in Europe and the United States. Piper methysticum extract is obtained by supercritical low temperature extraction of the dried rhizome of Piper methysticum. It can effectively inhibit the overreaction of sensory nerves, thus significantly improving the skin’s tolerance to external stimuli, and it is a natural cosmetic ingredient with strong analgesic and anti-irritant effects. Gentian Root Extract’s main components are gentiopicroside and amarogentin. It has the effect of clearing heat and removing dampness for traditional application. The main role of gentian root extract in cosmetics and skin care products is skin conditioner, odour inhibitor, with a lower risk factor. And it has a good activation effect on luciferase’s, which suggests that it has a role in atopic dermatitis and allergy. 2. Botanical Source
Kava, also named Piper methysticum Forst, is a perennial, erect shrubby medicinal plant of the Piperaceae family, native to the islands of the South Pacific, wild or cultivated, the roots and rhizomes are used as medicine, the active part being the fat-soluble resinous part. The main medicinal components of kava piper are carvacrolide, kawain. Kava pepper has long been a widely used medicinal ingredient in Europe and the USA for its skin calming, local anaesthetic, antibacterial and anti-anxiety effects, and is also widely used as a food supplement. The dried rhizome is used as a raw material for cosmetic products.
Gentian (Latin name: Gentiana scabra Bunge) is named for its leaves resembling nightshade and its taste resembling bitter gall. It is one of the oldest plants on earth, so it is known as “the living fossil of the plant kingdom” and one of the famous “four famous flowers on the plateau”. According to the records of “Compendium of Materia Medica”, Gentiana has the effects of “curing cold and heat between bones, frightening disease, inheriting injury, fixing the five internal organs, and killing insects and poisons”. Gentian mainly grows in North Korea, Japan, and Heilongjiang, Jilin, Liaoning, Zhejiang and other provinces in China. Gentian is a commonly used traditional Chinese medicine for a long time, only taking its root and rhizome. And in modern pharmacological research, gentian has been found to have a powerful effect in skin care.

4. PMSoothe® 6521 Function & Mechanism
- Nerve soothing effect
- Capsaicin receptor (TRPV1) protein content test
The main purpose of this test is to evaluate the soothing effect of PMSoothe® 6521 based on capsaicin-stimulated keratinocytes to detect TRPV1 protein content. TRPV1 is a gated ion channel receptor, also known as capsaicin receptor; it is a heat-sensitive receptor. After the action of capsaicin, etc., TRPV1 is activated, which further leads to Ca2+ influx. After adding PMSoothe® 6521, TRPV1 is inhibited, and the intracellular Ca2+ concentration decreases. Therefore, it indicates the test substance achieves a soothing effect by inhibiting TRPV1.

Based on keratinocytes, compared with the control group, the capsaicin receptor (TRPV1) protein content of sample PMSoothe® 6521 at a concentration of 0.03125% (m/v) decreased significantly, and the inhibition rate was 20.95%. It shows that the sample at this concentration under this test model, the soothing effect is achieved by inhibiting the expression of capsaicin receptor (TRPV1) protein.
- Never relieving test based on zebrafish
In this test, retinol was used to stimulate zebrafish to induce inflammatory pain in a larger area and for a longer period of time, resulting in increased movement trajectories and faster speeds. We use a behavior analyzer to detect the total movement distance and movement speed of zebrafish, and evaluate whether the sample PMSoothe® 6521 has a soothing effect. The result is as follows:

The results showed that the total movement distance of the 0.1% sample PMSoothe® 6521 was significantly reduced compared with the model control group, revealing that PMSoothe® 6521 had a soothing effect, and the soothing effect was 65%.

After PMSoothe®6521 was placed in different temperature conditions for 80 days, there was a little precipitation at high temperature, the color of the solution was slightly deepened. Under the other temperature conditions, the solution was clear and transparent, without precipitation, and had good- After PMSoothe®6521 was placed under different pH conditions for 80 days, there was a slight precipitation at pH 0 and pH 6.0, and the color of the solution was slightly deepened. At pH 9.0 and 10 there was a little precipitation. At the other pH conditions its water solution was clear and transparent, without precipitation, and had good stability.
- After PMSoothe®6521 was placed under the condition of butanediol for 80 days, there was a very trace precipitation of butanediol at high temperature, the color of the solution was slightly deepened. The rest butanediol conditions were clear and transparent, without precipitation and with good stability.

Formulation suggestions:

9. PMSoothe® 6521 Safety
- The “Announcement on the Name Catalog of Used Cosmetic Raw Materials” issued by the China Food and Drug Administration in 2014 and the 2010 version of the “International Standard Chinese Name Catalog of Cosmetic Raw Materials” of the China Fragrance Association both list Piper Methysticum Root Extract and Gentiana Scabra Root Extractas cosmetic raw materials.
- Plamed submitted PMSoothe®6521 for cytotoxicity testing, and the results showed that based on keratinocytes PMSoothe® 6521 showed no obvious cytotoxicity within the range of 0.3125mg/mL.
