Anti-inflammation is an eternal topic in the skincare industry: soothing, repairing, anti-aging, oil control and anti-acne products are all inseparable from anti-inflammation. These years, the mainstream anti-inflammatory ingredient in the is bisabolol (although it can be extracted naturally, most of the commercially available products come from lower-cost synthetic processes). Today we introduce a natural anti-inflammatory factor comparable to bisabolol: PMRelief® 9982.
PMRelief® 9982 is composed of peony root extract and Sophora flavescens root extract. The active ingredients are paeonol and matrine respectively. These two ingredients are well-known anti-inflammatory factors in the natural medicine field.
Sophora flavescens root extract contains various alkaloids, with matrine and oxymatrine as the main ones. It has strong anti-inflammatory and anti-allergic effects, can inhibit histamine and regulate the release of intracellular inflammatory mediators.
The main active ingredient of paeonia suffruticosa root extract is paeonol, which has pharmacological activities such as antibacterial and anti-inflammatory, anti-allergic, and immune function enhancing, and is widely used in cosmetics, pharmaceutical preparations and fine chemicals.

- Anti-inflammation effect based on zebrafish model
In this test, changes in the number of neutrophils in the skin of transgenic neutrophil green fluorescent strain of zebrafish (MPX) before and after treatment with Sodium Lauryl Sulfate (SLS) were used to test PMRelief® 9982’s soothing effects.
Use zebrafish within 2 days after fertilization for the experiment. One is that the mouth of the zebrafish has not yet developed well at this time, and SLS can only be absorbed through the skin; the other is that according to EU regulations, the zebrafish test at this time is not an animal experiment. Here is the test results as below.

The results showed that 0.05% of PMRelief® 9982 had a significant reduction in the number of neutrophils compared with the model control group, revealing its anti-inflammatory and soothing effect of 82%.
We also tested the number of neutrophils at the same concentration 0.0001% of PMRelief® 9982 and bisabolol (0.0001% is the maximum tolerated concentration of bisabolol) ), and the test results are shown below:

Under the present experimental conditions, the samples PMRelief® 9982 and Alpha-Bisabolol (95%) have the soothing efficacy (34% and 53% respectively), which are characterized by anti-inflammatory soothing efficacy.
- Skin soothing effect based on zebrafish’s movement distance
In this test, we stimulate zebrafish by glacial acetic acid that is strongly irritating and corrosive. Skin contact with glacial acetic acid will sting and burn. The zebrafish swims faster to get rid of the pain, and get out of the glacial acetic acid environment. Use behavioural analyser to test the total movement distance and the movement speed in zebrafish and to evaluate whether PMRelief® 9982 has a soothing effect.

It was observed that the total movement distance of PMRelief® 9982 was significantly reduced compared with the model control group, suggesting that the sample has the soothing efficacy.
- Anti-inflammation effect based on macrophagocyte
After the macrophages were stimulated with 1ug/mL LPS, the levels of pro-inflammatory cytokines (IL-1B, IL-6, IL-8, TNF-a) and inflammatory mediator (PGE2) were detected to evaluate the soothing effect of PMRelief® 9982.
PMRelief® 9982 showed a significant decrease in the content of pro-inflammatory factors (IL-1β, IL-6) and inflammatory mediators (PGE2) at a concentration of 0.03125% (m/v), with inhibition rates of 32.39%, 39.01% and 46.50%, respectively, indicating that it inhibited the secretion of pro-inflammatory factors (IL-1β, IL-6) and inflammatory mediators (PGE2) by achieving anti-inflammation efficacy.
What’s more, PMRelief® 9982 has no irritation according to the chick embryo chorioallantoic membrane experiment at concentration of 3%, and has no pesticide residues.
In conclusion, PMRelief® 9982 is of potent effect, high safety for skin and free of pesticide residues. It not only can relieve and soothe skin instantly, but also with great anti-inflammation property.
PMRelief® 9982 can be used in almost every type products, including lotion, serum, mask, cream and essence. Here are some tips for PMRelief® 9982 using in cosmetics formulation:
- The pH of 1% concentration of PMRelief®9982 is about 10, so it is recommended that dissolve PMRelief® 9982 in advance, and then adjust pH value to neutral condition (5.0-6.0), then add to the formulation.
- If there is a small amount of precipitation in low acid environments, we suggest to dissolve PMRelief®9982 with 5% butylene glycol.
- Under weak alkaline environment, PMRelief®9982 solution will becomes light yellowish brown color. Normal human skin pH is 4.5~6.5, and the pH of most skin care products is also weakly acidic, so the colour of PMRelief® 9982 solution will not change under acid or neutral condition.
