Before, Plamed test DCR®516’s anti-irritation effect based on capsaicin stimulation of keratinocytes. The test proved that DCR®516 can achieve the soothing skin effect by inhibiting the expression of TRPV1 protein.
So can DCR®516 continue to suppress the action potentials triggered by TRPV1 expression ?
In order to verify this effect, Plamed conducted a scientific experiment on the effect of DCR®516 on the action potential of primary cultured neuron cells. We are the first enterprise in China to conduct the experiment on the action potential and TRPV1 of kava extract.
DCR®516 Anti-irritation Test on Action Potential
This test was based on the anti-sensitivity mechanism that hinders nerve signal transmission, and used the whole-cell patch clamp technology to study the effect of DCR®516 on the action potential of primary cultured neuronal cells, and evaluated the effect on anti-sensitivity.
DCR®516 Anti-irritation Test Principle
After the rat DRG neuron cells form whole-cell seals, the recording mode is adjusted to current clamp; the current is clamped at 0pA, and the clamp current is increased from -30pA to 10pA for 1s, and then restored to 0pA to maintain 0.4 s.
In addition, the changes of drug efficacy under 1.5-2 times Rheobase stimulation were recorded, and the stimulation duration was 1s. The test data were acquired by an EPC-10 amplifier (HEKA) and stored in PatchMaster (HEKA).
DCR®516 Anti-irritation Test Result
The concentration-effect relationship of DCR®516 on the action potential of DRG neurons in rat suckling mice. The specific data are as follows:

DCR®516 can significantly reduce the release frequency of neuronal cell action potentials at the concentration of 3mg/mL and 10mg/mL;
DCR®516 had no significant effect on the action potential threshold (Threshold) at the concentration of 1mg/mL, 3mg/mL, and 10mg/mL;
DCR®516 had no significant effect on the peak amplitude at the concentration of 1mg/mL, 3mg/mL, and 10mg/mL;
DCR®516 at a concentration of 10mg/mL can significantly reduce the maximum depolarization rate of action potentials (Max up-stroke slope);
DCR®516 can significantly reduce the resting membrane potential (Membrane potential) at a concentration of 10 mg/mL;
DCR®516 had no significant effect on the stimulating current base intensity (Rheobase) at the concentration of 1mg/mL, 3mg/mL, and 10mg/mL;
DCR®516 can significantly reduce the membrane input resistance (The input resistance) at the concentration of 1mg/mL, 3mg/mL, and 10mg/mL.
DCR®516 Anti-irritation Test Conclusion
DCR®516 at a concentration of 1mg/mL can significantly reduce the the input resistance; while it had no effect on the number of spike, threshold, peak amplitude, max up-stroke slope, membrane potential and rheobase.
It can be seen from the above conclusion that DCR®516 can not only inhibit the expression of TRPV1 protein, but also continue to inhibit the action potential triggered by the expression of TRPV1, so as to inhibit the signal transduction after nerve cell excitation. It can be seen that DCR®516 can not only inhibit nerve excitation (inhibit TRPV1 protein expression) but also block nerve conduction after excitation (inhibit action potential) to play its anti-irritation role.
Generally, the nerve-relieving raw materials are only aimed at TRPV1 or only for nerve conduction, and DCR®516 has more efficacy advantages.
Plamed began to study kava extracts in 2018, and explored the influence of different extraction processes on the skin soothing effect of kava extracts. Finally, in 2020, we will find the best extraction process and launch the first generation of kava extracts with remarkable effects.
Other DCR®516 Anti-irritation Test
Zebrafish were treated with aspirin aqueous solution and DCR516 aqueous solution (24h), and then glacial acetic acid was used to establish a zebrafish pain model, and the effect of the product on zebrafish stimulation was judged by the total movement distance of the zebrafish.
The zebrafish test proves that the anti-irritant effect of DCR®516 is close to that of aspirin.
