1. PMRelief^® 9982 Overview

PMRelief^® 9982 is a compound of sophora flavescens root extract and paeonia suffruticosa root extract. Among them, sophora flavescens root extract contains various alkaloids (1%~2.5%), with matrine and oxymatrine as the main ones. Sophora flavescens root extract has strong anti-inflammatory and anti-allergic effects, which can inhibit histamine and regulate the release of intracellular inflammatory mediators, avoiding cells in an over-excited state and producing allergic and other inflammatory symptoms. It also has a strong inhibitory effect on hyaluronidase activity, thus preventing further damage to skin cells and providing excellent protection, and is an excellent immunosuppressant. The main active ingredient of paeonia suffruticosa root extract is paeonol, which has pharmacological activities such as antibacterial and anti-inflammatory, anti-allergic, and immune function enhancing, and is widely used in cosmetics, pharmaceutical preparations and fine chemicals. 2. Botanical Source

Sophora flavescens (Latin name: Sophora Flavescens Ait) is the dry roots of leguminous plant. It also can be called shrubby sophora. The plant is mainly located in hillsides, sandy land, grass slopes, shrubs and other places. It is a traditional Chinese herb plant. It is distributed in almost every province of North and South China. Also it exists in overseas countries, such as Russia, Japan, India, and North Korea. Sophora flavescens can clear away heat and dampness from human body; also kills insects and has diuretic therapy. It mainly has the pharmacological value of skin care, anti-tumor, anti-bacterial, anti-inflammatory and anti-arrhythmia.
Peony (Latin name: Paeonia Suffruticosa Andr.) is a deciduous perennial shrub of the family Paeoniaceae, mostly a cross between the purple-spotted peony and the Jizan peony, and is an important ornamental plant and one of the ten most famous flowers in China. Peony was once regarded as the national flower of China and was known as the “King of Flowers”, and because of its large and fragrant flowers, it was also known as the “national color and fragrance”. It is a precious flower unique to China and is native to the mountainous regions of the Qinling Mountains and the Daba Mountains in the west, with thousands of years of natural growth and more than 1,500 years of artificial cultivation.

• Zebrafish Anti-inflammationefficacy (neutrophil count) test

The action of SLS (Sodium laurylsulfonate) on the organism may trigger an irritant response in the organism. Irritant enters zebrafish to induce an inflammatory response, and neutrophils undergo an immune response, migrating and aggregating to the skin epidermis. Changes in the number of neutrophils in the skin of transgenic neutrophil green fluorescent strain of zebrafish (MPX) before and after treatment with SLS were used to test the sample for their soothing effects.

Phenotypes and bar graphs of the efficacy experiments for PMRelief^® 9982 are shown in Figures 1 and 2 below.

• Zebrafish soothing efficacy (total movementdistance) test

Although glacial acetic acid is a weak acid, it is strongly penetrating and corrosive. Skin contact with glacial acetic acid can cause a stinging and burning sensation. Zebrafish with underdeveloped swimming ability is selected and stimulated with glacial acetic acid to cause inflammatory pain over a larger area and for a longer period of time, resulting in more movement tracks and more pain. A behavioural analyser is applied to test the total distance travelled by the zebrafish and to evaluate whether PMSoothe^®9390 has a soothing effect. The results of PMRelief^® 9982 is tested as below.

• Inflammatory factor content assay (macrophage based) test

The main purpose of this experiment was to evaluate whether the substance to be tested has a certain soothing effect based on the change in the content of pro-inflammatory factors (IL-1β, IL-6) and inflammatory mediators (PGE2) detected by LPS-stimulated macrophages Raw264.7.

LPS (Lipopolysaccharide) is a component of the outer wall of Gram-negative bacterial cell wall, a substance composed of lipids and polysaccharides (glycolipids). The pathogenic mechanism of pathogenic bacteria is mainly the binding of LPS to Toll-like receptors, which activates the downstream inflammatory signaling pathway NFκB signaling pathway, NFκB is a nuclear transcription factor, which after activation enters the nucleus to bind to the promoter region of downstream target genes and initiate gene transcription, where the downstream target genes are mainly related cytokines and enzymes that mediate the inflammatory response. The synthesis of a large number of inflammatory factors makes the inflammatory response cascade amplified, further affecting the vascular response and inducing clinical phenomena such as skin erythema.

The test results of sample PMRelief^® 9982 are as follows.

PMRelief^® 9982 showed a significant decrease in the content of pro-inflammatory factors (IL-1β, IL-6) and inflammatory mediators (PGE2) at a concentration of 0.03125% (m/v), with inhibition rates of 32.39%, 39.01% and 46.50%, respectively, indicating that the sample inhibited the secretion of pro-inflammatory factors (IL-1β, IL-6) and inflammatory mediators (PGE2) by achieving soothing efficacy.
