Retinol and its derivatives are the members of vitamin A, it’s a natural compound with a variety of benefits. However, excessive use can cause skin irritation and allergic reactions. Select zebrafish with underdeveloped swimming ability and use retinol to stimulate zebrafish, which causes larger areas and longer periods of inflammatory pain, resulting in more movement trajectory and more severe pain. Using a behavior analyzer to detect the total movement distance of zebrafish to determine whether the sample has the soothing efficacy. | |
| Experimental
procedure |
1. Randomly select zebrafish in a 6-well plate with 15 tails per well.
2. Put the samples in water solution, and set up normal control group and model control group with a volume of 3 mL in each well 3. Incubate for 18h at 28 ℃, protected from light. 4. 10 zebrafish from each experimental group were randomly selected and transferred to a 96-well plate, and put retinol in water solution to establish the zebrafish pain model. Immediately use the behavior analyzer to determine the movement trajectory of zebrafish, analyze and collect the total movement distance (D) of zebrafish, and calculate the soothing efficacy of sample according to the formula to determine whether it has the soothing efficacy.

It was observed that the total movement distance of PMSoothe® 6521 was significantly reduced compared with the model control group, suggesting that the sample has the soothing efficacy.
Conclusion
Under the present experimental conditions, the sample PMSoothe® 6521 has the soothing efficacy, the soothing efficacy is 65%, which is specifically characterized by nerve soothing efficacy.
