Zebrafish Test of Natural Sensitive Skin Care Ingredient DCR516

DCR516 is widely applied in skin care products to smoothing sensitive skin. To prove natural cosmetic ingredient DCR 516’s safety, Plamed affords the test report to evaluate DCR516 relieving skin irritation on zebrafish.

1. DCR 516 Determine Standard

2. DCR 516 Test Method

2.1 Determine the maximum test concentration (MTC) of DCR516 to relieve sensitive skin

After each experimental group was treated for 24 hours, observe the toxic phenotype and count the number of deaths of each group zebrafish, and determine the MTC of DCR516’s effect to relieve skin irritation.

2.2 Evaluate the effect of DCR516 to relieve irritation on zebrafish

Randomly select 2 dpf normal wild type AB strain zebrafish in a 6-well plate, with 30 fish per well (each experimental group), and each well with a volume of 3 mL. DCR516 was tested at 2 concentrations: 222 and 667 μg/mL, and positive control drug aspirin is tested at 50 μg/mL, and a normal control group (zebrafish treated with fish water) and a model control group were set up at the same time. After each experimental group was treated for 24 hours, randomly selected 10 zebrafish from each experimental group and transferred them to a 96-well plate. One tail per well with a volume of 100 μL. Except for the normal control group, all the other experimental groups were given 0.009% glacial acetic acid to establish a zebrafish pain model, and a behavior analyzer was used to detect the total zebrafish movement distance (D) within 0.5 h. The total movement distance evaluates the irritation relieving effect of DCR516 on zebrafish. The irritation-relieving effect is calculated as follows:

3. DCR516 Test Result

Above all test results show that 0.67% DCR516 can significantly relieve skin irritation on zebrafish. So DCR 516 is an effective ingredient for sensitive skin care. It can be used with safety.
