2. Botanical Source
Kava pepper (Piper methysticum Forst) has been as edible plant for 3,000 years in the South Pacific islands, where it is considered “sacred” by the indigenous people and used in religious and cultural ceremonies. Kava is used to make kava wine, which is regarded as the national drink and is consumed when entertaining guests of honour, holding important festivals or discussing important matters. 4. PMRelief® 9292 Mechanism
- Anti-irritating
PMRelief® 9292 blocks the production of burning, pain and itching through two targets simultaneously, mainly acting on the skin sensory nervous system, which can both inhibit nerve excitation (inhibit TRPV1 protein expression) and block neurotransmitter transmission (inhibit action potential signaling), thus achieving the effect of neural soothing.
- Anti-inflammation
When the skin is exposed to external stimuli, a series of inflammatory cascades are triggered, resulting in the release of a variety of pro-inflammatory mediators, including biologically active amines (histamine and 5-hydroxytryptophan), cellular pro-inflammatory factors (IL-1 and TNF-α), inflammatory mediators and chemokines. The resulting vasodilation and increased vascular permeability allows leukocytes and plasma to extravasate to the site of injury or infection, leading to clinical inflammation such as redness of the skin. Neutrophils then undergo an immune response and migrate to the skin epidermis where they accumulate. Therefore, the anti-inflammatory effect of PMRelief® 9292 can be determined by measuring the number of neutrophils and the level of inflammatory factors.

- TRPV1 Protein Content Test
Based on keratinocytes, PMRelief® 9292 inhibited capsaicin receptor (TRPV1) by 43.29% and 50% at concentrations of 0.08% and 0.156%, respectively, which indicates PMRelief® 9292 inhibits the expression of TRPV1 protein in keratinocytes and has a soothing effect.
- Neural Signal Transmission Inhibition Test
Based on neuronal cells, PMRelief® 9292 inhibited the evoked frequency of neuronal action potentials (nerve signalling) by 92.86% at a tested concentration of 0.375 mg/mL.
- Inflammatory Factor Inhibition Test
Based on macrophage testing, PMRelief® 9292 inhibited TNF-α levels by 35.74% at 0.0625%, indicating anti-inflammatory efficacy.

- Test of Soothing Irritating Ingredients
Based on the chick embryo chorionic allantoic membrane test, 0.5% PMRelief® 9292 reduces the moderate irritation of salicylic acid to non-irritating; 0.5% PMRelief® 9292 reduces the moderate irritation of retinol to non-irritating; and 0.5% PMRelief® 9292 reduces the moderate irritation of 377 to non-irritating.
- Human Face Anti-Irritation Efficacy Test
The faces of 30 eligible subjects with sensitive skin were stimulated 0.001% capsaicin, and then after a single 30-min application of the product, subjects felt that the products provided 100% relief from burning, itching, and stinging, and 80% relief from redness of the skin.
6. PMRelief® 9292 Advantages
- Why Add Ginseng Extract?
Piper methysticum root extract is effective in soothing nerves, but not as effective in inhibiting inflammatory factors. However, ginseng extract showed a more prominent anti-inflammatory effect at a concentration of 0.002% based on neutrophil tests done on zebrafish.

PMRelief® 9292 solution of different pH under low temperature (4 ℃), room temperature (20-30 ℃) and high temperature (45 ℃) after 6 months, its aqueous solution in pH 7-9 conditions for a long time will produce a very small amount of precipitation; in the high temperature will be a little deepening of the colour. The rest of the conditions are clear and transparent, no precipitation, the product stability is good. Therefore, it is recommended to use PMRelief® 9292 at pH range of 4-6 and at room temperature.
