1. Overview
PCR® Zanthoxylum Bungeanum Extract is an active ingredient extracted from the pericarp of zanthoxylum bungeanum of the Brassicaceae family. Zanthoxylum bungeanum is China’s traditional medicine, spices, and flavoring agents, and are known as one of the “eight flavors” [1]. Native to China, its pericarps, seeds, stems and leaves are edible and medicinal. According to the Chinese Pharmacopoeia, zanthoxylum bungeanum has the effect of “warming the middle and relieving pain, killing worms and relieving itching” [2]. It’s used for epigastric cold pain, vomiting and diarrhea, abdominal pain, ascariasis; external treatment of eczema and itching. At present, with the in-depth study of the pharmacological effects of zanthoxylum bungeanum, the plant has a special effect of clearing away heat and cold, analgesic, spleen and other special effects, the medicinal components mainly include volatile oil, alkaloids, amides, lignans, coumarins and flavonoids. The active ingredients in zanthoxylum bungeanum have good pharmacological effects in the treatment of cardiovascular and digestive diseases as well as antioxidant pain [3]. In recent years, natural plant raw materials have been replacing chemically synthesized ingredients due to their advantages of low toxicity, high efficiency, degradation and low environmental pollution. 2. Botanical Source
Zanthoxylum bungeanum is a species of plant in the family Rutaceae. It is one of the sources of the spice Sichuan pepper. There are about 250 species of Sichuan pepper worldwide, widely distributed in tropical and subtropical regions of Asia, Africa, Oceania and North America. There are 39 species and 14 varieties in China [4], which are planted in both the north and south regions. Mainly produced in the northeast, Sichuan, Shaanxi, Shandong, Hebei and other places. The best quality is produced in Hanyuan, Sichuan, and is known as the “Da Hong Pao”. Mature fruits are harvested in the late summer, dried in the sun, and seeds and impurities are removed.

4. PCR® Zanthoxylum Bungeanum Extract Mechanism & Function
- Anti-itching
Plamed commissioned a third-party laboratory to evaluate the efficacy of the body lotion formulated with PCR® Zanthoxylum Bungeanum Extract in human beings for the purpose of relieving itchiness. The results of the test showed that PCR® Zanthoxylum Bungeanum Extract was highly effective in relieving itchiness in 100.0% of the test subjects. The test method and results are as follows:
Test method: The test adult mosquito (Aedes albopictus or Culex pipiens pallens) is placed in a test box, and the mosquito bites the volunteer’s arm and then applies the test body lotion (using the original dropper tip to draw up an appropriate amount and apply it directly and cover the discomfort completely, do not massage and rub). The other bites were placed on the skin without applying the medicine. The level of skin reaction to the bites was observed and recorded at regular intervals.
Soothing
- Anti-Inflammation Test
This test utilizes SLS to act on the genetically modified neutrophil green fluorescent strain zebrafish (MPX) may trigger the body to produce stimulus response, the stimulus enters the body of zebrafish, induces inflammatory response, neutrophils undergo an immune response, migrate to the epidermis of the skin and accumulate, through the change of the number of neutrophils in the skin to detect whether the sample PCR® Zanthoxylum Bungeanum Extract for its soothing effect. The results were as follows:
The results showed a significant reduction in the number of neutrophils in the sample PCR® Zanthoxylum Bungeanum Extract group at a concentration of 0.001% compared to the model control, revealing that the sample has an anti-inflammatory and soothing efficacy of 67%.
- Never Soothing Test
This test utilizes ice acetic acid to stimulate zebrafish to cause inflammatory pain over a larger area and for a longer period of time, resulting in increased trajectory and speed. We used a behavior analyzer to detect the total distance and speed of movement of zebrafish to evaluate whether the sample PCR® Zanthoxylum Bungeanum Extract has a soothing effect. The results are as follows:
The results showed that the zebrafish’s total movement distance traveled in the 0.001% PCR® Zanthoxylum Bungeanum Extract group was significantly reduced compared to the model control group, revealing that the sample had a nerve soothing efficacy of 28%.
- Human Capsaicin Patch Test
The experiment was divided into two groups: A and B. Group A was the blank control group, 0.5ml of 0.01% capsaicin were put on the patch test paper. Group B was PCR® Zanthoxylum Bungeanum Extract test group, 0.5ml of the prepared solution containing 0.01% capsaicin and 3% PCR® Zanthoxylum Bungeanum Extract were put on the paper. Then Groups A and B were put on the left and right arms at the same time. The patches were torn off after 30 min, and the results were as follows:
The results showed significant stinging, burning and redness in Group A, while no adverse effects were observed in Group B. The results of Group B showed no adverse effects. It was confirmed that the sample PCR® Zanthoxylum Bungeanum Extract could significantly improve skin tolerance and inhibit irritation caused by capsaicin.
- Anti-bacteria
Geng Zhi [5] determined the bacteriostatic activity by extracting the pericarps, seeds and leaves of Sichuan pepper and using the filter paper sheet diffusion method, using the size of the diameter of the inhibition circle as a measure. The results are shown in below table:
The results showed that the extract of zanthoxylum bungeanum pericarp had antibacterial effect on five kinds of test bacteria, and the antibacterial activity of the extracts from different parts of zanthoxylum bungeanum differed significantly, in which the antibacterial effect of the extract of zanthoxylum bungeanum pericarp was higher than that of the seeds and leaves. The antibacterial effect of zanthoxylum bungeanum pericarp extracts on the test bacteria was as follows: Staphylococcus aureus > Salmonella > Escherichia coli > Bacillus subtilis > Pseudomonas aeruginosa.

The results showed that the inhibitory effect of different parts of zanthoxylum bungeanum extracts on the isolated pathogenic bacteria varied greatly. Among them, the inhibitory effect of zanthoxylum bungeanum pericarp on pathogenic bacteria is outstanding, the inhibition rate can be controlled in more than 90%, the inhibitory effect on Aspergillus is particularly significant.
